摘要
目的观察黄芪建中汤对胃黏膜损伤模型大鼠基质金属蛋白酶-9(matrix metalloproteinase 9,MMP-9)及基质金属蛋白酶组织抑制剂-2(tissue inhibitors of matrix metalloproteinase 2,TIMP-2)表达水平的影响。方法将健康SD大鼠随机分为4组:假手术组、模型组、雷尼替丁组和黄芪建中汤组,每组10只。采用改良Okabe法复制动物模型。胃黏膜MMP-9、TIMP-2表达水平检测采用免疫组织化学法。结果模型组MMP-9表达较假手术组显著升高(P<0.01),各治疗组MMP-9表达水平显著低于模型组(P<0.01);模型组TIMP-2表达水平较假手术组显著降低(P<0.01),各治疗组TIMP-2表达水平显著高于模型组(P<0.01),黄芪建中汤组较雷尼替丁组TIMP-2表达水平显著升高(P<0.01)。结论黄芪建中汤通过调控MMP-9、TIMP-2的表达水平,修复胃黏膜损伤。
Objective To observe the effects of Huangqi Jianzhong (HQJZ) Decoction on the expression levels of matrix metalloproteinase 9 (MMP 9) and tissue inhibitors of matrix metalloproteinase 2 (TIMP-2)in rats with gastric mucosal injury. Methods Forty SD rats were randomly divided into 4 groups:Sham-operation group, model group, ranitidine treated group and HQJZ Decoction treated group. The model of gastric mucosal injury was induced by improved Okabe's method. The expression levels of MMP-9 and TIMP-2 of gastric mucosa were assayed by immunohistochemical method. Results The level of MMP-9 in model group was significantly higher than that in sham-operation group (P〈0.01),and in treatment groups it was significantly lower than that in model group (P〈0.01). The expression level of TIMP-2 in model group was significantly lower than that in sham-operation group (P〈0.01), while in treatment groups it was significantly higher than that in model group. The expression level of TIMP-2 in HQJZ Decoction treated group was significantly higher than that in ranitidine treated group(P〈0.01). Conclusion HQJZ Decoction repairs gastric mucosal injury by regulating the expression levels of MMP-9 and TIMP-2.
