摘要
目的研究以壳聚糖为骨架制备芦丁壳聚糖缓释片的工艺。方法以不同浓度的壳聚糖为辅料制备芦丁壳聚糖缓释片,用高效液相色谱法和释放度实验考察不同释放时间芦丁缓释片的溶出参数,并考察不同转速对释放度的影响。结果随着壳聚糖浓度的增加,芦丁释放速率变慢,1.5%壳聚糖醋酸溶液制备的芦丁片具有明显的缓释作用,其中位时间(t50)为(327.68±0.92)min,溶出63.2%所需时间(td)为(542.17±1.05)min,普通片t50为(4.93±1.03)min,td为(7.68±0.77)min,两者差异有极显著性(P<0.01);不同转速对释放度的影响主要在释放初期。结论壳聚糖对芦丁具有缓释作用,缓释效果良好,可作为芦丁缓释片的缓释辅料。
Objective To explore the technics for preparing sustained-release tablets of rutin containing chitosan as vehicle. Methods Different concentrations of chitosan were chosen as vehicles for preparation of sustained-release tablets.High performance liquid chromatography(HPLC) was used and a release rate was measured for them,in addition to the effect of different rotation speeds on the release rate. Results The release rate decreased along with the increase of concentrations of chitosan.It was showed that the rutin preparation had a good sustained release effect in 1.5% chitosan acetum.Compared with general tablets,t50 was(4.93±1.03) min,td was(7.68±0.77) min,the release parameters of sustained-release tablets were increased significantly(P0.01),t50 was(327.68±0.92) min,td was(542.17±1.05) min.The effect made by different rotations was significant at the beginning of drug release. Conclusion Chitosan slows the release of rutin from chitosan contained preparation.Therefore,chitosan can be used as the sustained release excipient for rutin sustained-release tablets.
