摘要
目的探讨芪连汤颗粒对糖尿病肾病(DN)大鼠过氧化物酶体增殖物激活受体-r(peroxisomeproliferators-activated receptor-r,PPAR-r)表达的影响。方法 SD大鼠55只,随机分为正常对照组10只,45只建立DN大鼠模型,并随机分为模型对照组、高剂量药物组,低剂量药物组,每组15只。高、低剂量药物组分别灌胃给予芪连汤颗粒溶液10,5 g.kg-1.d-1。正常对照组和模型对照组灌胃给予等量0.9%氯化钠溶液,连续4个月。取大鼠抗凝全血,分离单核细胞提取总蛋白,取动物肾脏组织总蛋白,用Western-blot方法检测PPAR-r与GAPDH比值(P/G)。结果正常对照组单核细胞内P/G值为(0.857 6±0.069 3),肾脏内P/G值为(0.912 6±0.066 7)。模型对照组单核细胞内P/G值为(0.621 9±0.095 0),肾脏内P/G值为(0.706 2±0.097 0)。高剂量药物组单核细胞内P/G值为(0.806 3±0.081 0),与模型对照组比较差异有显著性(P<0.05),肾脏内P/G值为(0.827 7±0.062 0),与模型对照组比较差异有显著性(P<0.05)。低剂量药物组单核细胞内P/G值为(0.764 8±0.087 0),与模型对照组比较差异有显著性(P<0.05),肾脏内P/G值为(0.785 7±0.099 0)。结论芪连汤颗粒能够保护DN大鼠,可能通过激活单核细胞系统和肾脏内PPAR-r表达,对DN大鼠的全身炎症状态进行调节。
Objective To explore the effect of qilian soup-granula on expressions of peroxisome proliferators-activated receptor-r（PPAR-r） in diabetic nephropathy（DN） rats. Methods 55 rats were divided into 10 normal rats and 45 DN rats,which subdivided into model control,high dose and low dose of qilian soup-granula,at 10 and 5 g·kg-1·d-1 for four months.Withdraw the whole blood and isolated monocytes（Mos）,and measured the protein expression of PPAR-r in both monocytes and kidneys with western-blot. Results The level of PPAR-r/GAPDH（P/G） in monocytes and kidney of normal control group were（0.857 6±0.069 3） and（0.912 6±0.0.066 7）,respectively;in model control were（0.621 9±0.095 0） and（0.706 2±0.097 0）;in qilian soup-granula high dose group were（0.806 3±0.081 0） and（0.827 7±0.062 0）,which were significant different from those in the model control（P0.05）.The level of P/G in low does of qilian soup-granula group was（0.7648±0.087）in Mos,which showed significant difference compaired with the model control. Conclusion Qilian soup-granula could protect against DN in rats,the mechanism of which may be related to eliciting express of PPAR-r in nephridial tissue and Mos,and adjusting the systemic inflammatory state on DN-rats.
