摘要
目的探讨干扰人胃癌细胞系BGC823胃泌素基因的表达后对细胞迁移能力的影响及其作用机制。方法采用Trans-weⅡ小室法检测稳定干扰胃泌素表达细胞株体外迁移能力的变化,采用Western blot技术检测干扰细胞株内MMP9蛋白表达的变化。结果胃泌素基因的表达下调使胃癌细胞穿过基质胶包被膜的数量显著下降,干扰胃泌素细胞内MMP9蛋白表达也显著降低。结论干扰胃泌素的表达可显著抑制胃癌细胞的迁移能力,这一作用可能通过下游MMP9基因的表达降低实现。
Objective To investigate the effect of BGC-823 cell's migration and its mechanism when interference of cell's gastrin gene expression Methods Migration capability of stably transfected cells in vitro was evaluated by using Transwell chamber model. Western blot was performed to detect the expression of MMP9 Results in BGC-823 cells which significantly down-regulate gastrin expression, the trans-membrane cell number decreased significantly and the expression of MMP9 is reduced also Conclusion Knock down of gastrin gene could suppress gastric cancer migration by inhibiting the expression of MMP9 protein
