摘要
为了解角质形成细胞作靶细胞进行基因转染、基因表达的可行性,用pSLXCMVβgal转染原代皮肤角质形成细胞,并观察其瞬时表达情况,发现转染后36小时细胞即可表达β半乳糖苷酶(βgal),表现为细胞蓝染,48小时蓝染细胞最多,约占总细胞数的15%。提示外源基因能够在角质形成细胞中有效地表达。
To investigate the possibility of keratinocytes as a target cell for gene transfection and gene expression, primary keratinocytes were transfected with pSLXCMV βgal and the transient expression of β galactosidase(β gal) was detected The results revealed that β gal was expressed in some cells 36h after transfection, and reached a high level at 48h after transfection with the percentage of positive cells of 1 5% Our results suggested that exogenous genes can effectively express in the keratinocytes
