实时荧光定量PCR检测大鼠肝癌组织中MEF2及ⅡHDACs的表达

Detection of MEF2 and Ⅱ HDACs Expressions in Rat Liver Cancer Tissue by Real-time Fluorescence Quantitative PCR

摘要 [目的]检测大鼠肝脏组织中MEF2A/2C mRNA及HDAC4/10 mRNA表达水平,并分析MEF2A/2C mRNA与HDAC4/10 mRNA表达的相关性。[方法]采用0.01%二乙基亚硝胺饲喂Wistar大鼠,建立大鼠肝癌模型,提取大鼠肝组织中的总RNA并逆转录为cDNA,应用Real-timePCR检测MEF2A/2C mRNA与HDAC4/10 mRNA表达量。[结果]18周后,大鼠致癌率为100%,总死亡率为35%,MEF2A/2C mRNA在第4、8周表达量显著增加(P<0.01),HDAC4/10mR-NA在第4周大量表达(P<0.01),随后都逐渐趋于正常水平。MEF2A/2C mRNA与HDAC4/10 mRNA表达水平在时间上不存在相关性。[结论]大鼠肝癌发生过程中,MEF2与肝癌组蛋白乙酰化有关,但MEF2A/2C mRNA与HDAC4/10 mRNA表达量在时间上无明显的关联性。 [Purpose] To investigate the expressions of MEF2A/2C and HDAC4/10 and the correlation between them in rat hepatocellular carcinoma. [Methods] Rat liver cancer model was established by feeding with 0.01% diethylinitrosamine (DEN) for Wistar rats. The total RNA isolated from rat liver tissue was reversely transcribed into cDNA, and the expressions of MEF2A/2C mRNA and HDAC4/10 mRNA was detected by real-time fluorescence quantitative PCR method. [Results] After 18 weeks, the carcinogenicity and mortality rate was 100% and 35%, respectively. The expression of MEF2A/2C mRNA was significantly increased at 4th and 8th week (P〈0.01), and HDAC4/10 mRNA increased at the 4th week(P〈0.01), and then all decreased gradually to normal level. There is a negative correlation between the expression of MEF2A/2C mRNA and HDAC4/10 on time regularity. [Conclusion] MEF2 plays an important role in histone acetylation in hepatocarcinogenesis. However, there is a negative correlation between the expression of MEF2A/2C and HDAC4/10 mRNA on time regularity during hepa- tocarcinogenesis in rats.
