摘要
构建过表达整合蛋白α５β１的ＳＭＭＣ７７２１细胞株。方法应用分子克隆技术和基因转染建立过表达整合蛋白α５β１的ＳＭＭＣ７７２１细胞株，应用ＲＴ－ＰＣＲ及流式细胞术鉴定ＳＭＭＣ７７２１细胞表面整合蛋白α５β１的表达。结果获得过表达整合蛋白α５／β１的ＳＭＭＣ７７２１细胞株α５．３－７７２１，β１．６－７７２１，α５β１．６－７７２１细胞。结论整合蛋白α５β１基因转染后，ＳＭＭＣ７７２１细胞整合蛋白α５β１的ＲＮＡ及蛋白质表达明显提高。
Purpose To establish SMMC7721 cell with line over expression of integrin α5β1. Methods Molecular clone technique,gene transfection,RTPCR and flow cytometry were used. Results We have established α5.37721, β1.67721 and α5β1.67721 cell lines over expressing integrin α5/β1. Conclusions Amount of integrin α5/β1 subunit increases on surface of SMMC7721 cells after transfection with integrin α5/β1 cDNA containing entire coding sequences.The establishment of these cell lines helps to study the functions of integrin α5/β1 in human hepatocellular carcinoma pathogenesis.
