摘要
目的通过研究腺嘌呤对下丘脑-垂体-肾上腺皮质轴的影响,阐明其导致"肾虚多尿"的机制。方法利用酶联免疫分析法分别检测正常组及腺嘌呤组血中促肾上腺皮质激素释放激素(CRH)、促肾上腺皮质激素(ACTH)、醛固酮(ALD)的含量;采用RT-PCR与免疫组化的方法检测垂体ACTHmRNA及蛋白的表达。结果与正常组比较,腺嘌呤可显著减少大鼠血中CRH、ACTH、ALD含量;并可下调垂体ACTHmRNA及蛋白的表达。结论腺嘌呤可影响HPA轴的功能导致水液代谢的功能障碍,研究结果从内分泌系统调节的角度阐明了腺嘌呤导致"肾虚多尿"的机制。
Objective To explain the mechanism of deficiency of the kidney and diuresis by discussing the influence of adenine on HPA.Methods The contents of CRH、ACTH、ALD in blood were determined,mRNA and protein expressions of ACTH with ELISA,RT-PCR and IHC technologies in model group and control group.ResultsCompared with control group,adenine could remarkedly decrease the contents of CRH、ACTH、cAMP in rat blood and mRNA and protein expressions of ACTH.ConclusionAdenine can decrease the contents of CRH、ACTH、ALD in rats blood and mRNA and protein expressions of ACTH by adjusting HPA.The results of the experiment expound the mechanism of deficiency of the kidney and diuresis adenine from HPA..
