高脂饮食诱导胰岛素抵抗大鼠血清apelin和visfatin水平变化及影响因素 被引量:1

Changes of Serum Apelin and Visfatin Levels in Rats with Insulin Resistance Induced by High-fat Diet and Its Influencing Factors

摘要 目的观察高脂饮食诱导的胰岛素抵抗(IR)大鼠血清apelin和visfatin水平变化,探讨apelin和vis-fatin与高胰岛素血症、体脂、糖脂代谢等的相关性。方法选择Wistar雄性大鼠30只,随机分为正常对照组(NC组,15只)和高脂饮食组(HF组,15只)。HF组大鼠喂以高脂饲料制备IR模型。喂养5周后行正葡萄糖-高胰岛素钳夹试验来确认IR大鼠造模成功后,测定两组大鼠血清apelin-36、visfatin、空腹胰岛素(FINS)、空腹血糖(FBG)、三酰甘油(TG)、总胆固醇(TC)水平及腹内脂肪含量、体质量。结果 HF组大鼠血清apelin-36、visfatin均显著高于NC组,差异有统计学意义(P<0.01)。相关分析显示:血清apelin与FINS、腹内脂肪含量、FBG、TC、TG、体质量呈正相关(r值分别为0.612、0.564、0.487、0.444、0.375和0.367,P<0.05);血清visfatin与FBG、FINS、腹内脂肪含量、TC、TG、体质量呈正相关(r值分别为0.781、0.756、0.747、0.592、0.521和0.297,P<0.05)。多元逐步回归分析显示,FINS是apelin的独立相关因素(R2=0.471,P<0.05);FINS和TC是visfatin的独立相关因素(R2FINS=0.407,P<0.01;RT2C=0.273,P<0.05)。结论 IR大鼠血清apelin和visfatin水平显著升高;血清apelin和visfatin与高胰岛素血症、体内脂肪含量、糖脂代谢相关;FINS是apelin和visfatin的独立相关因素,推测apelin和visfatin可能参与了IR的发生。 Objective To observe the changes of serum apelin and visfatin levels in rats with insulin resistance (IR) induced by high-fat diet to explore their correlation with hyperinsulinemia,body fat,glucose and lipid metabolism.Methods Thirty male Wistar rats were divided randomly into groups normal control (NC,n=15) and high-fat diet (HF,n=15).HF group were fed with high-fat diet to establish IR models.Hyperinsulinemia euglycemic clamp test was performed to confirm IR rat model establishment after 5 weeks.The levels of serum apelin-36,visfatin,fasting insulin (FINS),fasting blood glucose (FBG),triglyceride (TG),total cholesterol (TC) and intra-abdominal fat,body mass were determined in 2 groups.Results The levels of serum apelin-36 and visfatin were significantly higher in HF group than in NC group (P0.01).Correlation analysis showed that serum apelin was positively correlated with FINS,intra-abdominal fat content,FBG,TC,TG and body mass (r=0.612,0.564,0.487,0.444,0.3375,0.367,respectively),and that serum visfatin positively with FBG,FINS,intra-abdominal fat content,TC,TG and body mass (r= 0.781,0.756,0.747,0.592,0.521,0.297,respectively).Multiple stepwise regression analysis showed that FINS was an independent correlation factor of apelin (R2=0.471,P0.05);FINS and TC were independent correlation factors of visfatin (R2FINS=0.407,P0.01;R2TC=0.273,P0.05).Conclusion The levels of serum apelin and visfatin increase significantly in IR rats,serum apelin and visfatin are correlated with hyperinsulinemia,body fat content and lipid metabolism,and FINS is an independent factor of apelin and visfatin,suggesting that apelin and visfatin may be involved in IR occurrence.
