川芎嗪对大鼠局灶性脑缺血再灌注损伤的影响 被引量:23

摘要 目的观察川芎嗪对大鼠局灶性脑缺血再灌注损伤的影响。方法大鼠随机分为假手术组(Sham)、缺血再灌注组(IR)、川芎嗪组(TMP)。采用经大鼠右侧翼腭动脉线栓Willis环30min后再灌注的方法,建立大鼠局灶性脑缺血再灌注损伤的动物模型。动态观测局部脑血流(rCBF)及脑电图(EEG)的变化。测定血清丙二醛(MDA)、ATP酶(ATPase)、内源性超氧化物歧化酶(SOD)含量。结果与缺血再灌注组相比,川芎嗪组异常增高的MDA显著降低(P<0.05),血清和脑组织中SOD含量增加,ATP酶活性提高(P<0.05);川芎嗪组脑血流量(rCBF)提高,脑电图恢复至正常水平时程缩短。结论川芎嗪对局灶性脑缺血再灌注损伤大鼠有保护作用,其作用机制可能是减少大鼠局灶性脑缺血再灌注脂质过氧化、清除氧自由基、扩血管等作用。 Objective To explore the effect of teramethylpyrazine against cerebral ischemia-reperfusion injury in rats.Methods Rats were randomly divided into sham operation group(sham group),ischemia-reperfusion group(IR group),teramethylpyrazine group(TMP group).The focal cerebral ischemia reperfusion injury rat model was established by the suture occlusion of Willis ring through the right pterygopalatine segment of artery.The dynamic changes of regional cerebral blood flow(rCBF)and electroencephalogram(EEG)were observed.Serum malondialdehyde(MDA),ATP enzyme(ATPase),endogenous superoxide dismutase(SOD)levels were detected.Results Compared with IR group,the abnormal increase of MDA significantly reduced in TMP group(P〈0.05),SOD levels in serum and brain tissues and ATP activity increased significantly(P〈0.05),cerebral blood flow(rCBF)was improved,and the duration of EEG returning to the normal was shortened.Conclusion Teramethylpyrazine has an effect against focal cerebral ischemia-reperfusion injury,which may be induced by reducing focal cerebral ischemia and reperfusion lipid peroxidation,scavenging oxygen free radicals,and extending the blood vessels.
