甘氨酸对非酒精性脂肪肝大鼠血浆HMGB-1表达的影响及其意义 被引量:3

摘要 目的探讨甘氨酸对非酒精性脂肪肝(NAFLD)模型大鼠血浆中高迁移率族蛋白B-1(HMGB-1)表达的影响及其意义。方法雄性SD大鼠48只,随机分为3组,每组16只:对照组、模型组(高脂饮食)、治疗组(高脂饮食+甘氨酸)。造模第4、8周末分批处死大鼠。检测血浆肿瘤坏死因子-α(TNF-α)、谷丙转氨酶(ALT)、谷草转氨酶(AST)及内毒素(ET)、高迁移率蛋白B-1(HMGB-1)等指标。肝脏HE染色,观察肝脏的形态学变化。结果模型组大鼠4周已经出现脂肪性肝炎,8周炎症进一步加重,并出现轻微肝纤维化。与对照组相比,4、8周时模型组ALT、AST、TNF-α、ET、HMGB-1明显升高(P<0.05);治疗组上述指标较模型组均明显下降,差异有统计学意义(P<0.05)。结论在NAFLD中,HMGB-1可能以一定的机制参与了其炎症反应,进而影响了NAFLD的发生发展,甘氨酸有延缓非酒精性脂肪性肝炎炎症反应的作用。 Objective To investigate the effect of glycine on high mobility group box-1 protein(HMGB-1)expression in plasma and hepatic tissue homogenate in rats with nonalcoholic fatty liver disease(NAFLD)and its significance.Methods Forty-eight male SD rats were randomly divided into 3 groups:control group(normal diet),model group(high fat diet)and treatment group(high fat diet+glycine).All rats were sacrificed at the end of weeks 4,8.The levels of endotoxin(ET),alanine aminotransferase(ALT),aspartate aminotransferase(AST),tumor necrosis factor-α(TNF-α)and HMGB1 in liver were determined.The changes of hepatic morphology were observed by HE.Results The liver developed steatohepatitis in model group at the end of week 4.At week 8,the inflammation aggravated further and the liver developed mild hepatic fibrosis.Compared with control group,the levels of ALT,AST,TNF-α,ET and HMGB-1 in model group increased significantly at weeks 4,8(P〈0.05).The ALT,AST,TNF-α,ET and HMGB-1 levels in treatment group decreased compared with model group(P〈0.05).Conclusion In NAFLD,HMGB-1 may participate in the inflammation and then affect the occurrence and development of NAFLD.Glycine can effectively delay the inflammation of non-alcoholic steatohepatitis.
