摘要
[目的]为提高漆酶发酵水平提供一种有效的控制策略。[方法]采用栓菌菌株14001,对其发酵产漆酶条件及热激对漆酶酶活的影响进行研究。[结果]栓菌产酶最适Cu2+浓度为20mg/L,最适起始pH值为4.5,最适接种量为10%,最适培养温度为25℃,并对发酵过程进行pH控制,其酶活比不进行pH控制的酶活最大能提高30%;热激作用能诱导漆酶相应的mRNA产生,提高漆酶酶活1倍以上。[结论]优化后的培养条件和热激均能提高漆酶的发酵水平。
[Objective] The study aimed to provide an effective control strategy for enhanceing the level of laccase fermentation.[Method] With strain of trametes versicolor 14001,conditions for laccase production and the effects of heat-shock activity on fermentation was studied.[Result] The results indicated that the optimal fermentation conditions were as follows:Cu2+ concentration 20 mg/L,initial pH4.5 of the medium,inoculum percentage 10% and at 25 ℃.Enzyme activity improved 30% compared with not pH cortrol.Heat shock could induce the corresponding the mRNA of laccase,increased the laccase by one times.[Conclusion]The optimized performance and heat shock could enhance fermentation leverl of laccase.
