摘要
目的探讨苦参素防治心肌纤维化的效果。方法实验兔30只分为正常对照组、模型组及苦参素治疗组,每组10只,通过置于与脉冲发生器相连的电极建立兔心房颤动心肌纤维化模型。用图像分析系统测定兔左心房胶原分数、标准化血管周胶原面积及肌间动脉平均中膜厚度,采用RT-PCR法定量兔心肌组织转化生长因子β1mRNA的表达。结果经苦参素治疗后心房颤动心肌纤维化兔的左心房胶原分数、标准化血管周胶原面积及肌间动脉平均中膜厚度分别为0.057±0.029、0.82±0.33和(9.96±0.97)μm,低于模型组的0.112±0.047、1.75±0.48和(15.49±1.51)μm。兔心肌血管周围胶原及间质纤维化明显减少,心肌组织表达的转化生长因子β1mRNA降低了54.4%。结论苦参素具有一定的抗兔心肌纤维化作用,可能具有防治心肌纤维化的效果。
Aim To investigate the role of oxymatrine on myocardial fibrosis in rabbits with atrial fibrillation. Methods Experimental rabbits were divided into three groups: the control group,model group,and oxymatrine group. Myocardial fibrosis model of rabbits with atrial fibrillation was created by placing an electrode connected with a pulse instrument in the heart. Rabbit atrial collagen volume fraction,standardized perivascular collagen area,and average medial thickness were measured by automatic image analyzer. The expression of transforming growth faetorβ1 was determined by real-time quantitative RT-PCR. Results Atrial collagen volume fraction,standardized perivascular collagen area,and average medial thickness were 0.057±0.029,0.82±0.33, and (9.96±0.97)μm, respectively, for rabbits with myocardial fibrosis caused by atrial fibrillation treated with oxymatrine,all these were lower than that of model group (0.112±0.047,1.75±0.48,15.49±1.51). Expression of transforming growth factorial was also decreased 54.4% for rabbits group treated with oxymatrine. Conclusion Oxymatrine can improve myocardial fibrosis caused by atrial fibrillation in rabbits,it may protect the rabbits from development of myocardial fibrosis.
