摘要
文章从蚕沙中提取出叶绿素(CHL),并由此制备得到了蚕沙叶绿素铜配合物(Cu-CHL)。采用琼脂糖凝胶电泳实验研究了在白炽灯光照下Cu-CHL对pBR 322 DNA的切割能力。实验结果表明铜叶绿素对pBR 322 DNA具有光断裂能力,最佳断裂浓度范围为178.5~357μM。并采用紫外光谱、CD光谱和粘度法探讨了二者的结合模式,结果显示其为外部结合模式,结合常数为2.04×106m-1。
Copper chlorophyll(Cu-CHL) complex was prepared using Chlorophyll extracted from silkworm faeces. Agarose gel electrophoresis was carried out to investigate the photoc cleavage ability of Cu-CHL to pBR 322 DNA. The results suggested that Cu-CHL could induce DNA cleavage upon irradiation, and the optimization cleavage concentration range was 178.5-357 μM. Relative viscosity experiment, UV-Visible were used to study the interaction between Cu-CHL and ct-DNA. The results suggested that the binding mode of Cu-CHL and ct-DNA was perhaps the partly intercalative mode, the binding constant was 1.89x 106 M^-1.
