人骨髓基质细胞体外扩增方法的研究 被引量:1

摘要 目的为提高人骨髓成纤维细胞集落(CFU-F)的产率,缩短培养时间。方法在以往的人骨髓基质细胞体外扩增方法基础上进行了改进,每10ml培养体系中,含1×107个单个核细胞,以30%人AB血清代替小牛血清,增加了碱性成纤维细胞生长因子(bFGF),再加入氢化可的松和适量的IMDM培养液,培养第9d观察结果。结果在一定范围内人AB血清、bFGF均能促进CFU-F增殖,提高其产率,并缩短培养时间3~5d。结论可应用到人骨髓基质细胞体外扩增及传代中。 Objective Increasing the productivity of fibrocytic cell colony (CFU-F) formed by human bone marrow and shortening the time for the culture.Methods The available method for proliferating human bome marrow stromal cells in vitro was improved.In the culture system,there are 1×10 7 monocytes in each 10 ml.Thirty percent of AB serum are used in place of calf serum,then the basic fibrocytic growth factor(bFGF) is added to the culture system with subsequent addition of hydrocortisone and proper amoumt of IMDM.The result is observed on the nineth day after the culture.Results Human AB serum and bFGF can promote CFU-F,increasing productivity and shortening the time of the culture by 3 to 5 days.Conclusion This method can be used for proliferating human bone marrow stromal cells in vitro and it is of signficance to the study of the character of CFU-F and the application to hematopoietic disease.
