摘要
目的:以三文鱼皮为原料,采用中性蛋白酶制备胶原肽,研究其抗氧化活性。方法:在单因素试验的基础上,利用正交试验优化工艺条件;用凝胶渗透色谱法测定酶解液分子质量分布;测定胶原肽成品清除2,2-二苯基-1-苦基肼(DPPH)自由基能力和还原力,评价其抗氧化活性。结果:最优酶解条件:酶量0.15%(以鱼皮计),鱼皮-水质量比1:3.0,酶解温度55℃,pH6.0,酶解时间6h。在此条件下,胶原肽的最高得率74.28%。酶解液中分子质量在180~1000Da之间的肽占88.08%。胶原肽成品具有较强的清除DPPH自由基能力,IC50为1.302mg/mL,同时也表现出较强的还原力,其中2.5mg/mL胶原肽的还原力是相同浓度BHT的0.69倍。结论:由本法所得胶原肽是以分子质量180~1000Da之间的寡肽(约2~8个氨基酸残基)为主的物质,具有较好的抗氧化活性。
Objective:Using salmon skin as raw material,the peptides from collasen has been prepared by neutrase and its antioxidant activity was also investigated.Methods:On the basis of single factor tests,the technological parameters of prepareation were determined by orthogonal test.The molecular weight of the hydrolysate was determined by GPC.And its antioxidant activity was evaluated by determining the scavenging activity on 2,2-diphenyl-1-picrylhydrazyl(DPPH)radical and the reducing power.Results:The optimumed condition were as follows:amount of neutrase was 0.15%(based on salmon skin),the ratio of skin-to-water was 1:3.0,temperature was 55 ℃,the pH value was 6.0,the time of hydrolysis was 6h,under this condition the maximum yield of peptides was 74.28%.The molecular weight of the hydrolysate was determined and 88.08% the total was in the range of 180 to 1 000 Da.The peptides obtained showed the stronger scavenging activity on DPPH radical,IC50 value was 1.302 mg/mL.And its reducing power was 69 percent of BHT at 2.50 mg/ml.Conclusion:The peptides obtained is mainly composed of peptide with 2 to 8 amino acid residues,and has good antioxidant activity.
