摘要
目的观察染料木素(GEN)对异丙肾上腺素所致大鼠心肌肥厚的保护作用及其对心肌组织抗氧化能力的影响。方法雄性SD大鼠48只,随机分为对照组,心肌肥厚模型组,溶剂组,低、中、高剂量GEN组。造模成功后第2天开始给予不同浓度的GEN、二甲基亚砜或生理盐水,连续14 d,末次给药后禁食12 h,取心脏称左心室质量,计算左心室质量参数,比色法测定心肌组织超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性及丙二醛(MDA)含量,酶法测定血清及心肌组织乳酸脱氢酶(LDH)和肌酸激酶(CK)活性。结果心肌肥厚模型组左心室质量参数升高,心肌组织SOD和GSH-Px活性降低、MDA含量升高,血清LDH和CK活性升高,而心肌组织LDH和CK活性降低。GEN治疗组与心肌肥厚模型组相比各项指标均明显改善(P<0.05)。结论 GEN可通过提高SOD和GSH-Px活性,提高心肌组织抗氧化能力,减少心肌酶的释放,从而起抗心肌肥厚作用。
Objective To observe the protective effect of Genistein(GEN) on rat myocardial hypertrophy caused by isoproterenol.Methods The myocardial hypertrophy was made using subcutaneous injection with isoproterenol at the site of rat back,the dose was 1 mg/(kg·d) and it continued for 10 days.The rats with myocardial hypertrophy were divided into six groups randomly and treated with various dose GEN,DMSO and normal saline respectively for 14 days.The left ventricle index was measured,the activity of superoxide dismutase(SOD) and glutathione peroxidase(GSH-Px) and the content of malondiadehyde(MDA) were observed by colorimetric method.The serum or myocardial tissue enzymes activity of lactate dehydrogenase(LDH) and creatine kinase(CK) were evaluated by enzymic method.Results The control groups treated with DMSO or normal saline indicated that the left ventricle index,elevated enzyme activity in LDH,CK at serum and MDA level at myocardial tissue increased,the activities of SOD,GSH-Px,LDH and CK at myocardial tissue decreased.The rats with myocardial hypertrophy treated with GEN showed that left ventricle index,MDA level in myocardial tissue,enzyme activities of LDH and CK in serum decreased(P〈0.05).Conclusions GEN has a protective effect on the myocardial hypertrophy caused by isoproterenol and its mechanism associates with promoting antioxidation.
