程序性死亡因子配体2在不同类型冠心病患者外周血单核细胞中的表达

摘要 目的探讨不同类型冠心病患者外周血单核细胞上程序性死亡因子配体2(CD273/PD-L2)的蛋白以及mRNA表达水平。方法将81例行冠状动脉造影的患者根据造影结果及临床表现分为急性冠脉综合征(ACS)组36例,稳定型心绞痛(SAP)组25例,对照组20例。流式细胞仪检测外周血单核细胞CD273+细胞百分率,RT-PCR检测其mRNA表达。结果与对照组比较,SAP及ACS组患者外周血单核细胞上CD273蛋白及mRNA表达均升高,以ACS组为著(P<0.01)。结论冠心病患者外周血单核细胞CD273蛋白和mRNA表达水平明显升高。 Objective To explore the expressions of the protein and mRNA of programmed death factor ligand 2(CD273/PD-L2) in peripheral blood monocytes of different types of coronary artery disease.Methods Eighty-one patients undergoing coronary angiography were divided into three subgroups: acute coronary syndrome(ACS)(n=36),stable angina pectoris(SAP)(n=25) and control patients(n=20).The mononuclear cells were acquired through Ficoll density centrifugation,CD14^+CD273^+ protein and CD273 mRNA expression levels were determined by flowcytometry and RT-PCR.Results Comparing with the control,the levels of protein and mRNA expression of CD273 in peripheral blood monocytes of SAP and ACS increased,especially in the ACS group(P〈0.01).Conclusions The expressions of protein and mRNA of CD273 in CD14^+ peripheral blood monocytes of coronary artery disease patients were significantly higher than that of the control group.
