摘要
根据已发表的猪圆环病毒2型(PCV2)基因序列,分段设计2对PCV2特异性引物,用PCR方法扩增近年从福建省部分发病猪场分离鉴定的7株PCV2的全基因序列并进行比对分析.结果表明:7株PCV2基因组全长1767 bp,与GenBank上发表的17株PCV2参考毒株的同源性为94.7%-99.5%;7株PCV2分离株之间的全基因同源性高达98.4%-99.5%;7株PCV2 ORF2编码蛋白均具有很强的亲水性和抗原性,其核苷酸及推导的氨基酸同源性高达98.9%-99.7%和97.9%-99.6%,与参考毒株的同源性分别为91.1%-99.7%和85%-100%,存在一定的差异.可见,7株PCV2在基因水平上差异不显著,在地域分布上也没有明显差异.
According to the published gene sequence of procine circovirus type 2(PCV2) in GenBank,two pairs of specific primers were designed and synthesized to amplify the complete genome of seven PCV2 strains isolated from local epidemic pig farms in Fujian province.The complete genome sequences of the seven PCV2 were compared with published genomes of seventeen PCV2 strains in GenBank.The results showed that the genomes of the seven PCV2 strains were 1767 bp in length.The homologous of the complete genomic nucleotides among the seven isolates and compared with 17 PCV2 strains in GenBank was 98.4%-99.5% and 94.7%-99.5% respectively.The ORF2 genes of the seven isolates were provided with highly hydrophilic property and immunogenicity.The homologous of nucleotide sequence and amino acid of the ORF2 genes were 98.9%-99.7% and 97.9%-99.6% among the seven isolates respectively,and were 91.1%99.7% and 85%-100% compared with the PCV2 strains in GenBank.The results indicated that complete genomes of the seven PCV2 strains were highly homologous and no significant differences were observed in genetic and geographic origin.
