摘要
目的:通过研究皮肤切创愈合过程中血管内皮生长因子(Vascular endothelial cell growth factor,VEGF)、转化生长因子β1(Trans forming growth factor-β1,TGF-β1)的表达与损伤时间的关系,探讨VEGF、TGF-β1能否在法医学判断损伤时间方面作为一个指标。方法:用大鼠建立皮肤切创模型,按随机原则分为实验组、正常对照组和死后损伤组,采用免疫组化(Immunohi stochemistry,IHC)方法对大鼠皮肤切创愈合过程中VEGF、TGF-β1的表达变化进行观察。结果:实验组VEGF、TGF-β1在创伤周围组织中的表达高于对照组及死后伤组(P<0.05);VEGF在创伤后3~24h主要在中性粒细胞、巨噬细胞和成纤维细胞中表达,阳性表达的强度与创伤后时间正相关;2d出现在新生肉芽组织血管内皮细胞、巨噬细胞及成纤维细胞中,呈强阳性表达,于伤后4d达到峰值;以后各组阳性细胞逐渐减少,呈弱阳性表达,与对照组比较差异有显著性(P<0.05);TGF-β1的生前伤后变化规律与VEGF基本相似。结论:VEGF、TGF-β1在创伤修复过程中的表达呈现时序性规律,可以为法医学损伤时间推断提供一种新的指标。
Objective:To investigate whether the expression of protein VEGF and TGF-β1 can be used as an indextoestimate injuryage in the field of forensic medicine by approaching the temporal relation of VEGF and TGF-β1 expression with injury in the healing process of the incised wounds on rat skin.Methods:Experimental rat models were established by incisions made on the rat skin.the rat models were divided randomly into three groups:the experimental group,the normal group,and the postmortem injured group.The expression of VEGF and TGF-β1 were observed with Immunohistochemical techniques in their process of cicatrization.Results:In the experimental group,the expression of VEGF and TGF-β1 were higher than those in the normal control and postmortem injury group(P0.05).The expression of VEGF mainly existed in the neutrophilic granulocyte,macrophages and fibroblasts 3~24 h after injury,and its positive level increased in direct proportion to the wound age.Two days after the injury,the presence of VEGF was observed in the macrophages, fibroblasts,and the vascular endothelial cell of neoformative granulation tissue with strong positive expression,reaching a summit on the 4thday after the injury.The positive cells in both groups then gradually decreased,with the protein expression turning weakly positive. Their difference with the normal control group is significant(P0.05).The changing rule of TGF-β1 expression in the experimental group was similar to that of the VEGF expression.Conclusion:The expression of VEGF and TGF-β1 during the cicatrization process follows temporal rules,thus can be provided as an new index to judge the Injury age in the field of forensic medicine.
