人骨髓间充质干细胞体外定向诱导成骨细胞的研究 被引量:7

摘要 目的为建立一种简便有效的下颌骨缺损修复治疗的新方法——利用体外分离纯化人骨髓间充质干细胞(marrowstromal cells,MSCs),诱导向成骨细胞转化后再用于临床。方法 Ficoll密度梯度离心后贴壁培养法,分离纯化MSCs体外培养,经连续传代培养,加入10-7mol/L地塞米松,10-2mol/Lβ-甘油酸钠,500 ug/L的维生素C诱导液。结果 MSCs诱导后细胞形态向成骨细胞转化,经RT-PCR、流式细胞仪、细胞碱性磷酸酶活性、细胞糖原染色鉴定为成骨细胞。结论 hMSCs经定向诱导显示成骨细胞特性,为临床下颌骨缺损组织工程修复提供种子细胞。 Objective To discover a new method of mandibular reconstruction after surgical ablation,and to induce human mesenchymal stem cells into osteoblasts in vitro for the clinical use of tissue engineering bone.Methods Mesenchymal stem cells(MSCs) were isolated by lymphocyte separatiom medium,and were cultured in vitro.Serial passaged cells were fed with conditioned medium.Osteoblast was induced by 10-7mol/L decaesadril,10-2mol/Lβ-sodium glycerophosphate,500ug/L anticorbic acid.Results The MSCs of human cultured grew and survived well in vitro.After induction,MSCs turned into osteoblasts.Passaged cells were examined by RT-PCR,flow cytometry,alkalinephosphatase(ALP)and andcellglycogen.Conclusions The results demonstrated that human mesenchymal stem cells from bone marrow and umbilical cord blood could be induced to differentiate into osteoblasts in vitro.They could be used as seed cells for bone tissue engineering of mandibular construction.
