Livin基因表达与食管鳞癌化疗药物敏感性的相关性研究

摘要 目的研究原代培养食管癌细胞对化疗药物的敏感性,并进一步探讨其敏感性与食管癌组织中Livin基因表达的关系,为食管癌的临床化疗提供个体化用药的实验资料和依据。方法:取手术切除的新鲜食管癌组织标本,用MTT法检测经化疗药物作用后肿瘤细胞活力及代谢活性的变化;同时取相同食管癌组织制成石蜡切片,用免疫组化检测Livin蛋白的表达。结果:1.Livin阳性表达与5-FU及ADM耐药有关(P<0.05);2.TAX、MMC及5-FU与ADM及DDP之间敏感性差异具有统计学意义(P<0.05),前者明显高于后者;ADM敏感性明显高于DDP,组间差异具有统计学意义(P<0.01);结论:食管癌组织中Livin阳性表达者与5-FU及ADM耐药有关,提示Livin可作为食管癌原发性耐药标志之一。 Objective:study to evaluate the chemosensitivity of primary culturing Esophageal carcinoma cells by using MTT and the protein expressions of Livin in Esophageal carcinoma tissues,and to explore the relationship between Livin expression and chemosensitivity of Esophageal carcinoma.We also determined whether these results can be used to help therapy the Esophageal carcinoma.Methods: Inhibitory rate of cells was detected by MTT assay;Expression of Livin in Esophageal carcinoma tissues were detected by immunohistochemistry(IHC).Results: 1.The positively Livin stained cells showed resistance to 5-FU and DDP.2.the sensitivity of MMC,5-FU and TAX was significantly higher than DDP and ADM(P0.05);In the later,the sensitivity of DDP was significantly lower than ADM(P0.01);Conclusion: The Livin positive cells showed resistance to 5-FU and ADM.Our results suggest that Livin positive expression may contribute to primary drug-resistance of Esophageal carcinoma.
