儿茶素对超氧阴离子自由基诱导下血管内皮祖细胞TERT表达的影响

Effect of Catechin on Telomerase Reverse Transcriptase Expression in Endothelial Progenitor Cells Induced by Superoxide Anion

摘要 目的研究儿茶素对氧自由基诱导的血管内皮祖细胞(endothelial progenitor cells,EPCs)端粒酶逆转录酶基因(telomerasereverse transcriptase,TERT)表达的影响。方法分离培养Sprague Dawley大鼠骨髓内皮祖细胞。黄嘌呤-黄嘌呤氧化酶系统生成外源性超氧阴离子自由基(O_2^-)。化学发光法检测不同浓度(5.0μg/mL、7.5μg/mL、10.0μg/mL、12.5 μg/mL、15.0μg/mL)儿茶素对O_2^-的清除效果:TUNEL法检测内皮祖细胞凋亡;RT-PCR检测TERTmRNA表达;Western-blot检测TERT蛋白表达。结果儿茶素对O_2^-的清除效果呈浓度剂量效应。结论儿茶素可降低O_2^-对内皮祖细胞凋亡增加、TERT mRNA与蛋白表达下降的影响。 Objective To study the effect of catechin on telomerase reverse transcriptase (TERT) expression in endothelial progenitor cells induced by oxygen free radicals. Methods The endothelial progenitor cells of the Sprague Dawley rat bone marrow are isolated and cultured. Xanthine-xanthine oxidase system is used to generale exogenous superoxide anion ( O^-2 ). Chemil are used to test the effect of catechin with different concentrations (5.0 μg/mL,7.5 μg / mL, 10.0 μg / mL, 12.5 μg / mL, 15. 0 μg / mL) on removing the O^-2 , TUNEL for the apoptosis of endothelial progenitor cells, RT-PCR for TERT mRNA expression, Western-blot for TERT protein expresion level. Results catechin removes O^-2 in a dose-dependent manner. Conclusion The increased apoptosis of endothelial progenitor cells and decreased TERT mRNA and protein expression level due to O^-2 will be both lowed by cateehin.
