摘要
研究了培养条件和诱导条件等对工程菌CH1的最终菌体量和表达量的影响,确立最优的高密度发酵条件,还探讨了不同补料流加方式对菌体生长的影响。结果表明,DO-stat流加方式具有显著增加菌体量的作用,发酵液最终菌体密度D600达25.31,湿菌体53.4 g/L,蛋白表达量也达到32.3%,实现了ChIFN-γ的高密度发酵和高表达。初步纯化的鸡γ干扰素以500 U/只的剂量对双A肉鸡口服给药,可以提高机体的巨嗜细胞吞噬活性(P<0.05)。本试验为鸡γ干扰素的工业化生产和临床应用提供了依据。
Establishing a procedure for the high cell density fermentation and the effect on the chiken macrophages activity of a recombinant ChIFN-γ.Several factors including cultivation conditions and induction conditions of strain CH1 that influenced the final cell density and promoting ChIFN-γ production was optimized.The effects of three feeding strategies were also investigated.The result showed that DO-stat feeding strategy had significant effect in enhancing final cell density.Under the established conditions,the final cell dencity reached D60025.31 and more than 53.4 g/L of wet bacteria per liter could be obtained.The expression level of aim protain was higher than 32.3%.Chiken macrophages activity was highly increased under oral doses of 500 U per double A dorking(P0.05).The study provides a basis for the industrial production and clinical application of ChIFN-γ.
