摘要
目的:测定阿替洛尔片的血药浓度,对健康壮年、老年及肾病志愿者进行药物动力学研究。方法:健康壮年及老年志愿受试者各8 例,肾病志愿者5 例,均为单剂量口服阿替洛尔片75 mg ,用 H P L C 法测定。结果:药—时曲线符合一级吸收二室模型。健康壮年人的主要药动学参数为 Ka :1 .01 h - 1 , T1/2β:4 .28 h , K10 :0 .25 h - 1 , A U C:5 .2 μg·h·ml- 1 , C L/ F:14 .95 mg·ml·(μg·h) - 1 , Tmax :2 .38 h , Cmax :0 .73 μg·ml- 1 。健康老年人主要药动学参数为 Ka :1 .17 h - 1 , T1/2β:8 .51 h , K10 :0 .17 h - 1 , A U C:7 .56 μg·h·ml- 1 , C L/ F:8 .94 mg·ml·(μg·h) - 1 , Tmax :2 .72 h , Cmax :0 .73 μg·ml- 1 。肾病患者主要药动学参数为: Ka :1 .02 h - 1 , T1/2β:45 .28 h , K10 :0 .09 h - 1 , A U C:15 .29 μg·h·ml- 1 , C L/ F:5 .02 mg·ml·(μg·h) - 1 , Tm?
OBJECTIVE:Pharmacokinetics of atenolol tablets were studied in 8 young healthy volunteers、8 old healthy volunteers and 5 nephrotic patients after oral single does administration of 75 mg.METHODS:serum atenolol concentration were determined by HPLC.RESULTS:The serum concentration time profiles were fitted to two compartment model with first order absorption.CONCLUSIONS: T 1/2β 、 K 10 、 AUC and CL/F are distinctly different among the youth,the old and the nephrotic patients ( P < 0.01 ).
