摘要
在以前研究工作的基础上,我们用抗人尿Tamm-Horsfall蛋白(THP)抗体包被聚苯乙烯酶标板建立酶联免疫吸附法(ELISA),其标准曲线范围3.1～100ng/ml,灵敏度2ng/ml与放射免疫测定法(RIA)一致。实验内变异系数4.2±3.3％,实验间变异系数9.5±5.1％。36例正常人血清THP为51.5±130.75ng/ml。55例慢性肾炎、尿毒症、肾结石及肝硬化病人血清THP,除慢性肾炎外与正常人相比差异均有显著性(P<0.05和0.005)。15例肾结石病人尿THP比正常人增高(P<0.005)。
An enzyme-linked immunosorbent assay (ELISA) has been established using polystyrene immunoplates coated with antibody to human Tamm-Horsfall urinary glycoprotein (THP) on the basis of our previous work. The standard curves showed linearity between 3.1—100 ng/ml with a sensitivity of 2 ng/ml and the method was consistent with radioimmuno-assay. The intra-assay and inter-assay coefficients of variation were 4.2±3.3% and 9.4±5.1% respectively. The serum THP in 36 normal subjects was 51.51±13.57 ng/ml. In comparison with normal value, the serum THP values of 22 nephrolithiasis and 10 uremia patients were significantly lower, those of 7 cirrhotics were higher and those of 14 chronic nephritis patients showed no significant difference. Urinary THP in 15 nephrolithiasis cases was much higher than normal.
