摘要
本文报道用人肝癌细胞株SMMC-LINM建立带瘤裸鼠模型,以此研究抗人小扁豆凝集素结合型甲胎蛋白异质体单克隆抗体(AFP-R-LCAMcAb)VG5的肿瘤显象能力和规律。结果提示,肝癌组织能够选择性地聚积McAbVG5,在一定时限内其摄取量随时间延长逐渐增加。96小时肿瘤组织与肝脏和脾脏的摄取率比值分别为5.27和5.16,肿瘤定位指数为5.16。γ照相显示肿瘤部位有明显的放射性积聚,可得到较清晰的肿瘤显象。
VG5 is one of murine monoclonal antibodies against human AFP-R-LCA. In vitro VG5 had a high specific binding capacity with human hepatic cancer tissue and cell lines. ^(131)Ⅰ-labeled VG5 and ^(125)Ⅰ-labeled normal murine IgG were injected into nude mice bearing human hepatoma cell line SMMC-LINM in order to compare the differences between VG5 and IgG distributions in tumor and various normal tissues. The distribution of radioactivity in vivo suggested that ^(131)Ⅰ-VG5 had good localization in hapatic cancer. The tumor/liver and tumor/spleen ratios were 5.27 and 5.16 respectively. The lccalization index of tumor was 5.16 at 96 hr. The radio activity of ^(125)Ⅰ-NMIgG in tumor was much lower than that of ^(131)Ⅰ-VG5. The results suggest that monoclonal antibody VG5 could be used in imaging and treatment of hepatic cancer.
