摘要
目的:以大鼠肺Ⅱ型细胞为靶细胞,检测烹调油烟的细胞毒作用和对细胞DNA的损伤。方法:烹调油烟颗粒提取物对大鼠肺Ⅱ型细胞染毒24h后,应用MTT法、溴乙锭荧光法以及单细胞凝胶电泳技术检测烹调油烟对大鼠肺Ⅱ型细胞的细胞毒性和DNA的损伤。结果:在受试剂量范围内,烹调油烟颗粒提取物具有细胞毒性,可致DNA交联及断裂的作用随染毒剂量的增加而增强,并存在剂量-反应关系(r细胞毒性=-0.948,P<0.01;r交联=0.976,P<0.01;r断裂=0.957,P<0.01)。结论:烹调油烟颗粒提取物对大鼠肺Ⅱ型细胞具有细胞毒性作用和DNA损伤作用,可使DNA链间交联和DNA单链断裂。
bjective: The purpose of the study was to detect the cytotoxicity and
DNA damage in rat pulmonary type II cells induced by cooking oil fume. Methods: Type II lung
cells were isolated from rat pulmonary tissues and after being cultured for 24 hours,they were
exposed to acetone extracts from vegetable oil fume(oil temperature:270).Cytotoxicity and DNA
interstrand crosslinks as well as DNA single strand breaks were detected by MTT assay,
ethidium bromide fluorescence assay and the single cell gel electrophoresis assay. Results:
The extracts had an obvious cytotoxicity and DNA damage effect to the cells and
doseresponsive relationships were found (rcytotoxicity=-0.948,P<0.01;rDNA
crosslinks=0.976,P<0.01;rDNA breaks=0.957,P<0.01). Conclusion: Cooking oil fume has a
cytotoxicity and genotoxicity to rat pulmonary cells, which may be a molecular basis for
carcinogenesis of cooking oil fume.
