血脉健抗动脉粥样硬化家兔氧化损伤的研究

摘要 目的:探讨血脉健对食饵性动脉粥样硬化家兔氧化损伤的改善作用.方法:将健康日本大白耳兔50只,随机分出10只为空白对照组;其余40只通过免疫刺激和喂饲高脂饲料复制动脉粥样硬化模型,按血脂水平随机分为模型组、阿托伐他汀组、血脉健组,每组各10只.除空白对照组和模型组灌胃冷开水外,阿托伐他汀组灌胃阿托伐他汀混悬液,每天0.45 mg/kg,血脉健组灌胃血脉健药液0.4 g/kg,连续灌胃1月.于末次灌胃后2 h兔耳中动脉取血,分离血清测定血清超氧化物歧化酶(SOD)活性、丙二醛(MDA)和氧化低密度脂蛋白(ox-LDL)含量,取脑组织匀浆测定SOD活性和MDA含量.结果:与空白对照组比较,模型组血清和脑组织MDA、血清ox-LDL含量显著升高(P〈0.01),血清和脑组织SOD活性显著降低(P〈0.01);与模型组比较,血脉健组和阿托伐他汀组血清SOD活性和脑组织总SOD活性升高,血清和脑组织MDA含量、血清ox-LDL含量降低,差异有统计学意义(P〈0.01).血脉健组和阿托伐他汀组血清MDA、ox-LDL含量明显高于空白对照组(P〈0.05、P〈0.01),而脑组织MDA含量略高于空白对照组,但无统计学意义(P>0.05),血清SOD活性与空白对照组基本一致,但脑组织总SOD活性明显低于空白对照组(P〈0.05).血脉健组和阿托伐他汀组之间SOD活性、MDA、ox-LDL含量基本一致,差异无统计学意义(P>0.05).结论:血脉健能显著提升血液和脑组织SOD活性,减少MDA和血清ox-LDL的含量,具有较好的对抗动脉粥样硬化家兔氧化损伤的作用. Objective: To investigate 'the blood and health of the prey of atherosclerosis in rabbits to improve the role of oxidative damage. Methods: Healthy Japanese Pak 50 rabbits, 10 rats were ceded to the control group; the remaining 40 by immune stimulation and copy high fat diet fed atherosclerosis model, according to lipid levels were randomly divided into model group, A simvastatin group care, blood health group, n=10. In addition to the control group and model group was given the cold water, the atorvastatin group was given atorvastatin suspension, daily 0.45 mg/kg, blood blood Kin Kin group was given liquid 0.4 g/kg, continuous administered in January. At 2 h after the last administration of rabbit ear artery to take blood samples of serum superoxide dismutase (SOD) activity, malondialdehyde (MDA) and oxidized low density lipoprotein (ox-LDL) levels, and brain tissues homogenate SOD activity and MDA content determination. Results: Compared with the control group, model group, serum and brain tissue MDA, serum ox-LDL was significantly higher (P〈0.01), serum and brain tissue SOD activity was significantly decreased (P〈0.01); with model group, blood health group and atorvastatin serum SOD activity and total SOD activity increased in brain tissue, serum and brain tissue MDA, serum ox-LDL levels decreased significantly (P〈0.01). Healthy blood group and atorvastatin serum MDA, ox-LDL was significantly higher than the control group (P〈0.05, P〈0.01), while the brain tissue MDA content was slightly higher than the control group, but no statistical significance (P〉0.05), serum SOD activity consistent with the control group, but brain tissue SOD activity was significantly lower than control group (P〈 0.05). Healthy blood group and between the atorvastatin group SOD activity, MDA, ox-LDL levels consistent with, the difference was not statistically significant (P〉0.05). Concolusion: The blood vessels can significantly improve the health of blood and brain tissue SOD activity and reduction of serum MDA and ox-LDL levels, with better atherosclerosis in rabbits against oxidative injury.
