酶联免疫印迹法对弓形虫速殖子功能性抗原分析

摘要 研究弓形虫功能性抗原,为开发弓形虫病诊断试剂及疫苗提供理论基础。方法:采用十二烷基硫 酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE)和免疫印迹方法,对弓形虫抗原及弓形虫速殖子抗原中能与免疫兔血清和 弓形虫抗体阳性人血清起特异性反应的抗原组分进行分析。结果:弓形虫速殖子抗原的蛋白组分在SDS-PAGE中 用125 g/L的分离胶可产生较好的分辨率,分离出 30多条区带。免疫印迹分析表明 2种抗血清均能特异地识别弓 形虫速殖子抗原,共同识别的抗原组分相对分子质量为 43 000、33 000、27 000、14 000。结论:这些抗原组分是诱导 宿主产生对弓形虫感染免疫应答的功能性抗原。 Aim:To provide theoretical basis for developing the diagnosticum and vaccine through the analysis of tachyzoite antigens of toxoplasma gondii. Methods:Using SDS-PAGE and immunoblot analysed the antigens and tachzoite antigens of toxoplasma gondii that can react on the antisera of rabbits and human specifically.Results:the SDS-PAGE patterns of antigens of tosoplasma gondii consisted of about 30 polypeptide bands.The better resolution of these antigens was obtained by using 125g/L polyacrylamide gels. Thebands of 43000,33000,27000and 14000were commonly recognized with antisera of huaman and rabbit Conclusion:The four major bands of proteins were considered to be functional antigens that induced host to respond to toxoplasma gondii, suggesting they may be the candidate antigens for the vaccine reserch of toxoplasma gondii and diagnosis of Toxoplasmosis.
