猪血浆蛋白粉中IgG含量的酶联免疫检测方法研究 被引量:2

摘要 猪血浆蛋白粉已经成为断奶仔猪理想和必需的蛋白饲料来源。评价血浆蛋白粉质量的重要指标就是其中IgG的含量。研究通过制备猪IgG单克隆抗体,调试优化后建立了相应的酶联免疫检测方法。该方法最低检测浓度为10μg/g,阴性血浆蛋白粉添加10(1%)、100(10%)、200mg/g(20%)猪IgG标准品后用本方法测定回收率为73.1%~104.6%,批内、批间变异系数小于15%,可基本满足血浆蛋白粉质量监控需要。 Spray-dried Plasma Protein (SDPP) has now become ideal weaned piglets' protein supplement, and immunoglobulin G is the most important factor that expresses the quality of it. In the current research, an enzyme linked immunoassay method for the detection of IgG in SDPP was developed based on a specific monoclonal antibody against pig IgG. The detection limit of the method is 10 μg/g, and SDPP samples free of IgG spiked with IgG standard at the concentrations of 10 (1%), 100 (10%), 200 mg/g (20%) indicated that the recovery of the method were between 73.1% and 104.6%, the intra-assay, inter-assay C.V. were both less than 15%, the applicability of the method was then demonstrated.
