摘要
目的:建立提取植物产品中γ-氨基丁酸的前处理方法。方法:筛选最佳提取方法,并对该法提取过程中的一些因素(磺基水杨酸的浓度、水浴震荡时间、温度、浸提时间等)进行优化,确定了用浓度为5%磺基水杨酸,在30℃热水浴中震荡15 min后,静置12 h的提取方法,最后于氨基酸自动分析仪进行分析定量。结果:实验表明磺基水杨酸法在内标实验中的回收率可达98.48%,比盐酸水解法提高13.54%~13.77%,比乙醇回流-水提法提高7.42%~8.06%。优化提取方法样品的平均加标回收率为94.40%~99.81%。相对标准偏差(RSD)为0.23%。结论:本方法回收率高、重现性好,是简易可靠的前处理技术方案。
Objective:To establish the pretreatment method for extraction in γ-aminobutyric acid of plant products.Methods:Screening of the best extraction method,and we made further research on some facts in the extraction process(the concentration of sulfosalicylic acid,shock time in water bath,temperature,extraction time,ect.).Identified the extraction method that are with the concentration of 5% sulfosalicylic acid,in hot water bath,standing 12 h,and finally in automatic amino acid analyzer to analyze quantitative.Results:The sulfosalicylic acid method,including the recovery rate was up to 98.48% in internal standards,compared with hydrochloric acid hydrolysis increased 13.54%~13.77%,compared with ethanol extraction by 7.42%~8.06%.Optimization of sample extraction,the average recovery was 94.4%~99.81%.The relative standard deviation(RSD) was 0.23%.Conclusion:The method of high recovery and good reproductbility,is simple and reliable preprocessing program.
