摘要
目的制备戊型肝炎病毒诊断基因芯片并进行杂交验证。方法利用Oligo6.0软件针对戊型肝炎病毒诊断基因保守区域设计多对寡核苷酸探针,用芯片点样仪将其按照阵列设计打印在玻片上制备成基因芯片。样品标记采用限制性显示技术,样品标记后进行杂交,进而在芯片扫描仪中对得到的探针数据进行分析并实验验证。结果芯片杂交后得到的探针荧光强度好,信噪比高,符合临床样品的基因亚型,RT-PCR验证后得到的电泳结果与芯片实验一致。结论实验设计并制备的寡核苷酸戊型肝炎病毒诊断芯片能用于戊肝的检测,为这项疾病的实验室诊断提供了一种快速平行的检测方法。
Objective To study the preparation of diagnostic DNA microarray for hepatitis E virus(HEV).Methods The specific primers of oligo probes were designed with the Oligo 6.0 program according to the conserved region of HEV.The DNA microarrays were prepared by spotting the probes to the surface of glass slides by the robotics.Restriction display PCR was used to label the samples.Results The results showed that the oligo probes were specific hybridized with HEV samples.And from the hybridizing signals on microarray,the specificity and sensitivity in the HEV diagnosis were satisfactory and the results were also detected by RT-PCR.Conclusion Using the probes to prepare the DNA microarray for clinical diagnosis of HEV is a quick and effective method.
