免疫酶标DNA探针对肿瘤坏死因子mRNA表达的原位杂交分析

摘要 采用磺基化修饰的TNF DNA探针,在原位杂交水平上,以免疫酶反应为显色系统,研究了内毒素对正常人单核细胞 TNF mRNA的诱导作用。结果表明,阳性细胞胞浆内呈细颗粒聚集的染色,围绕胞核分布。诱导的 TNF mRNA 高峰为刺激后3~4小时。而上清液中 TNF的活性高峰则为10小时左右。此外,仅部分单核细胞呈现TNF mRNA的转录,提示可能存在产生TNF的单核细胞亚群。 The induction of TNF-mRNA in human monocyteg by endotoxin was detected after in-situ hybridization using a sulfonated TNF DNA probe with development by immunoenzymatic approach (APAAP). The results showed that positive monocytes were stained in cytoplasm with small particles scattered around the nucleus. The peak of TNF-mRNA induced occurred 3 to 4 hours after exposure to endotoxin, but maximal release of TNF in supernatanta occurred at about 10 hours. In addition, only part of monocytes showed the transcription of TNF-mRNA, suggestiog the possibility of the presence of TNF-producing monocyte subsets.
