牡丹叶片愈伤组织诱导过程生化指标研究

摘要 采用牡丹(Paeonia suffruticosa)试管苗的叶片作为外植体,配制两个不同激素浓度的诱导培养基组合,研究牡丹叶片愈伤组织形成过程中可溶性糖、可溶性蛋白、超氧化物歧化酶(SOD)、过氧化物酶(POD)的含量以及活性的变化规律。结果表明,在最初的7~14d,生长素浓度较低的诱导培养基MS+2,4-D1.0mg/L+6-BA0.5mg/L+NAA1.0mg/L的外植体可溶性糖含量表现为缓慢下降,而生长素浓度较高的诱导培养基MS+2,4-D2.0mg/L+6-BA0.5mg/L+NAA2.0mg/L的外植体表现为缓慢上升;14d以后两种诱导培养基中外植体可溶性糖的含量均表现为先上升,21d时达到峰值,接着下降;两种诱导培养基外植体可溶性蛋白含量的变化规律表现为迅速上升、迅速下降、缓慢下降,培养到14d时达到峰值;SOD活性变化规律与可溶性糖相似;两种诱导培养基的外植体POD活性在前28d一直下降,28d后培养基MS+2,4-D1.0mg/L+6-BA0.5mg/L+NAA1.0mg/L的外植体POD活性逐渐上升,培养基MS+2,4-D2.0mg/L+6-BA0.5mg/L+NAA2.0mg/L则继续下降。 Two induction media with different hormone combinations were designed to study the change rule of soluble sugar content, soluble protein content, SOD activity and POD activity during callus induction of tree peony (Paeonia suffruticosa) leaf in vitro.The results showed that soluble sugar content of explants cultured on induction medium with low auxin concentration (MS+2,4-D 1.0 mg/ L+6-BA 0.5 mg/L+NAA 1.0 mg/L) decreased slowly while that cultured on induction medium with high auxin concentration (MS+2,4-D 2.0 mg/L+6-BA 0.5 mg/L+NAA 2.0 mg/L) gave the reverse trend during the initial 7~14 days of culture.After 14 days, soluble sugar content of explants cultured on two induction media both increased, and reached a peak value at 21 days, then decreased.Soluble protein content of explants cultured on two induction media both gave a change trend of rising quickly, decreasing quickly, then decreasing slowly, and reached a peak value at 14 days.SOD activity of explants cultured on two induction media gave the same change trend as that of soluble sugar content.POD activity of explants cultured on two induction media both decreased in the fore 28 days, after which POD activity of explants cultured on MS +2,4-D 1.0 mg/L+6-BA 0.5 mg/L+NAA 1.0 mg/L increased slowly, while that cultured on MS +2,4-D 2.0 mg/L+6-BA 0.5 mg/L+NAA 2.0 mg/L still decreased.
