摘要
采用高效液相色谱法对诱导产生的籼稻粤泰B(YTB)成熟胚愈伤组织继代培养过程中内源多胺组分和含量进行了测定。结果表明采用XDB-C18柱流动相梯度(从40%甲醇到80%甲醇)洗脱可以使Put、Spd和Spm3种多胺组分得到很好的分离。水稻粤泰B成熟胚愈伤组织继代培养5d后体内Put和Spd的含量明显增加,而Spm的含量基本不变。推测Put和Spd对水稻愈伤组织的生长发育可能有重要的作用,可为优化水稻愈伤组织培养基的组分提供参考。
The kinds and contents of endogenous polyamines in callus from matured embryo of indica rice YTB durin subculturing were determined by HPLC in this paper.The results showed that 3 polyamines, including putrescine (Put) spermidine (Spd) and spermine (Spm) were well separated on XDB-C18 column with a mobile phase of grads (from 40% methanol to 80% methanol).The contents of Put and Spd in the callus from matured embryo at 5 d after subculturing wer obviously increased than those at 1 d, but no obvious change in the content of Spm was observed.This suggested that Pu and Spd might had some effects on the growth and development of rice callus, which provided a reference for optimizing th composition of the substrate for culturing rice callus.
