糖尿病和肥胖症患者网膜脂肪组织中的内脂素表达及其影响因素 被引量:1

Expression of visfatin in visceral adipose tissue and its influential factor in type 2 diabetics and obese patients

摘要 目的探讨糖尿病、肥胖症患者网膜脂肪组织细胞内脂素的表达及其影响因素。方法选择糖尿病组、肥胖症组、健康对照组各60例,测定血清内脂素、脂联素、肿瘤坏死因子α(TNF-α)浓度。同时,选取三组共66例(糖尿病组23例、肥胖症组22例、对照组21例)行外科手术时切割的各内脏器官网膜脂肪组织提取总mRNA,然后进行Northern b lot印迹技术研究,分析内脂素mRNA差异表达以及与其他脂肪因子的相关关系。结果糖尿病和肥胖症患者血清内脂素浓度较对照组明显升高(P均<0.01),糖尿病、肥胖症血清内脂素浓度与TNF-α呈正相关(r=0.441,P=0.045;r=0.541,P=0.037)。网膜脂肪组织中内脂素mRNA的灰度值与血清内脂素、TNF-α水平呈正相关(r=0.771,P=0.021;r=0.500,P=0.041)。结论内脂素在2型糖尿病内脏脂肪及血清中高表达,TNF-α可能促进内脂素表达。 Objective To investigate the expression of visfatin in visceral adipose tissue in type 2 diabetics and obese patients,and the influential factor of visfatin.Methods The levels of plasma visfatin,adiponectin,TNF-α and other clinical parameters were assayed in all 180 subjects(including 60 type 2 diabetics,60 obese patients and 60 controls).Visfatin mRNA was abstracted in visceral asipose tissue in different internal organ of 66 subjects(including 23 type 2 diabetics patients,22 obese patients and 21 controls)during surgery.And then Northern Blot test was used.The correlation between visfatin mRNA and other fat factors was analyzed.Results The levels of plasma visfatin in type 2 diabetics and obese patients were higher than that of the control group(all P0.01).The levels of plasma visfatin in type 2 diabetics and obese patients were correlative positively with the concentration of TNF-α(r=0.441,P=0.045;r=0.541,P=0.037).The expression of visfatin mRNA in visceral adipose tissue was correlative positively with the concentrations of plasma visfatin and TNF-α(r=0.771,P=0.021;r=0.500,P=0.041).Conclusions Visfatin in visceral adipose tissue and plasma in type 2 diabetes mellitus expresses very strongly,TNF-α may promote the expression of visfatin.
