山羊GnRHR基因部分序列多态性及其与产羔数的关联性分析 被引量:9

摘要 采用聚合酶链式反应-单链构象多态性(PCR-SSCP)技术,用3对引物鉴定了黄淮山羊和波尔山羊促性腺激素释放激素受体(GnRHR)基因外显子2和外显子1部分序列的多态性,并对黄淮山羊Gn-RHR基因多态性与产羔数的关联性进行了分析。结果表明,引物P1、P3扩增片段在2种山羊中均只检测到1种基因型,不存在多态性;而引物P2扩增片段,2种山羊类型均检测到AA、AB和BB基因型。黄淮山羊AA、AB和BB基因型频率分别为0.725、0.200和0.075;波尔山羊AA、AB和BB基因型频率分别为0.765、0.206和0.029。测序分析发现AA型基因与BB型基因相比在58bp处发生A→C碱基突变,该突变导致编码氨基酸由赖氨酸改变为谷氨酰胺。在黄淮山羊中,BB型比AA型个体平均产羔数多0.66只(P<0.05)。因此,山羊GnRHR基因第1外显子具有多态性;GnRHR基因可能是控制黄淮山羊多胎性能的一个主效基因或是与之存在紧密连锁的一个遗传标记。 The polymorphism of exon 1,2 of estrogen receptor (GnRHR) gene in Huanghuai goat and Boer goat were detected by PCR-SSCP method and the relevance between the polymorphism of GnRHR gene and the litter size in Huanghuai goat were analysed. The results indicated that there was one genotype(AA)detected by primer 1 and 3 in two goat breeds,and three genotypes (AA ,BB and AB)in Huanghuai goat and Boer goat detected by primer 2. In Huanghual goat,the frequencies of AA,BB and AB genotypes were 0. 725,0. 200 and 0. 075, respectively. In Boer goat, the frequeneies of AA, BB and AB genotypes were 0. 765,0. 206 and 0. 029 respectively. Sequencing revealed one mutation (58A→C) of exon 1 in genotype BB compared with genotype AA, which gave rise to amino acid changes(Lys→Gln). The litter size in the Huanghuai goat with mutant homozygous genotype BB was 0. 66(P〈0. 05) larger than that with wild type AA. Thus, exon 1 of GnRHR gene in goat has polymorphism GnRHR gene may beone of the main effective gene controlling the prolificacy of goat or a genetic marker linked with it closely.
