淋巴细胞Fas和配体(FasL)表达变化及其与抗肿瘤活性的关系 被引量:8

THE RELATIONSHIP BETWEEN THE EXPRESSION OF Fas AND FasL ON LYMPHOCYTES AND ITS ANTI TUMOR ACTIVITY

摘要 为探讨淋巴细胞表面Fas及其配体(FasL)在IL-2和IL-12作用下表达的变化,以及Fas和FasL与淋巴细胞抗肿瘤活性的关系,作者用免疫荧光标记和流式细胞仪分析了IL-2和IL-12激活的杀伤细胞(IL-2AK和IL-12AK)Fas和FasL的表达;分别用乳酸脱氢酶(LDH)释放法和annexin-V-FITC标记检测IL-2AK和IL-12AK杀伤的肿瘤细胞的坏死和凋亡。结果表明:①正常人外周血淋巴细胞(PBL)(20.40±5.69)%表达Fas,但无FasL表达;3株消化道肿瘤细胞系恒定表达Fas,始终不表达FasL;②IL-2与IL-12可诱导LAK细胞表达FasL(P<0.01),并促使Fas表达显著增加(P<0.01);③LAK细胞致肿瘤细胞死亡中FasL表达与annexin-V标记阳性率和肿瘤细胞表面Fas表达正相关。由此推论,Fas-FasL途径是IL-2与IL-12诱导活化的LAK细胞杀伤肿瘤的途径之一,淋巴细胞致肿瘤凋亡分别与淋巴细胞FasL表达和肿瘤细胞Fas表达一致。 To determine the expression of Fas and Fas ligand (FasLor CD95L) on lymphocytes which were activated by IL 2 and IL 12, and to bring light the relationship between their expression and the anti tumor activity of the lymphocytes. Fas and FasL were labeled with immunofluorescence, and determined by flow cytometer. LDH release and annexin V Fluo /PI labeling were used, respectively, to detect the necrosis and the apoptosis of tumor cells. The results showed that ①Lymphocytes from normal donors carried (20.4±5.69)% of Fas and nor FasL, three tumor cell lines expressed constantly Fas but no FasL;②IL 2 and IL 12 could induce the expression of FasL and significantly increased the Fas expression on LAK cells;③The FasL expression was positively correlated with the positive rate of annexin V labeling and Fas expression on tumor cells killed by the LAK cells. It was concluded that the expression of FasL on IL 2AK and IL 12AK and the Fas on tumor cells had positive correlation with the apoptosis of tumor cells; and Fas and FasL was one of pathways that IL 2AK and IL 12AK killed tumor targets.
