摘要
目的:探讨凉血通瘀方治疗急性出血性中风的机制。方法:采集并制备急性出血性中风患者的血清,检测NO浓度和NOS的活性;建立体外血管内皮细胞H2O2氧化损伤模型,检测细胞上清NO浓度和NOS的活性。结果:凉血通瘀方作用于急性出血性中风患者后其血清NO水平明显升高(P<0.05),凉血通瘀方作用于H2O2氧化损伤的血管内皮细胞其细胞上清NO、NOS水平明显升高(P<0.05)。结论:凉血通瘀方可在一定程度上增强血管内皮细胞NOS活性,促进NO的分泌,这可能是凉血通瘀方治疗急性出血性中风的作用机理之一。
Objective:To explore the molecular mechanism of liangxue tongyu fang in the treatment of acute hemorrhagic stroke.Methods:Serum of patients with acute hemorrhagic stroke was collected,VECs were cultured in vitro,an injured model by H2O2 was established,and the NO concentration and activity of NOS were detected by NO and NOS kits in different groups.Results:The results showed that under the effects of liangxue tongyu fang on the patients with acute hemorrhagic stroke,the NO levels in serum were significantly elevated (P0.05),and under the effects of the clinical equivalent amount of liangxue tongyu fang on vascular endothelial cells(VECs)damaged by H2O2,NO,NOS levels were significantly elevated (P0.05).Conclusion:To some extent,Liangxue Tongyu could increase the activity of NOS to promote the secretion of NO.This may be one of the molecular mechanism of liangxue tongyu fang in the treatment of acute hemorrhagic stroke.
