摘要
目的探讨G蛋白偶联受体激酶2(GRK2)在猪血清诱导的免疫性肝纤维化模型中表达的变化及其意义。方法 Wistar大鼠腹腔注射猪血清建立免疫性肝纤维化模型,在造模的第0、3、6、9、12、16周不同时间点采用Masson染色对肝脏组织作病理检查;免疫组化染色法、蛋白免疫印迹法检测肝脏中GRK2表达水平的变化。结果大鼠腹腔注射猪血清在16周时可以成功建立免疫性肝纤维化模型。与正常大鼠相比,免疫性肝纤维化大鼠肝脏中GRK2的表达量随造模时间的延长而明显降低。结论免疫性肝纤维化大鼠肝脏中GRK2表达水平下调,提示GRK2可能在免疫性肝纤维化过程中发挥重要作用。
Objective To investigate the variation of GRK2 expression in porcine serum-induced liver fibrosis rats.Methods Wistar rats were intraperitoneally injected with 0.5 ml of porcine serum twice a week to establish immunological liver fibrosis model.Masson stain was used to examine the hisopathological change in time course of immunological liver fibrosis model(0,3,6,9,12,16 week).The expression of GRK2 was detected by Western blot analysis and immunohistochemistry staining.Results The immunological liver fibrosis model in rats was induced successfully at the end of the 16 th week after the injection of porcine serum.In the early stage,the GRK2 expression was significantly intensity,while gradually decreased before the injection was stopped.Conclusion The GRK2 expression of rats liver decreases in porcine serum-induced hepatic fibrosis,which indicates that GRK2 may play an important role in the process of immunological liver fibrosis.
