摘要
目的观察通络益肾温阳方对DN大鼠肾组织α-SMA、TNF-α表达的影响,从抗肾纤维化角度探讨该方的作用机理。方法采用单侧肾切除术+腹腔注射STZ复制DN大鼠模型。分为正常对照组、病理对照组、通络益肾温阳方治疗组、洛汀新+糖适平治疗组,观察各组大鼠肾组织α-SMA、TNF-α的表达及肾脏的病理改变。结果通络益肾温阳方治疗组大鼠肾组织α-SMA、TNF-α的表达显著低于病理对照组(P<0.01),肾组织病理损害均较病理对照组明显减轻。结论通络益肾温阳方对DN大鼠有一定的治疗效果,其机理可能是通过抑制肾组织α-SMA、TNF-α表达,减轻肾间质纤维化程度来实现的。
Objective To observe the effect of Tong Luo Yi Shen Wen Yang formula on kidney tissue＇s α-SMA and TNF-α in rats with diabetic nephropathy,and to probe into the mechanism of the formula from the aspect of anti-renal-fibrosis.Methods DN rats model was made by unilateral nephrectomy ＋ intraperitoneal injection of STZ. The rats were divided into normal control group,pathological control group,Tong Luo Yi Shen Wen Yang formula group,and Lotensin ＋ Gliguidone group. The expression of kidney tissue＇sα-SMA and TNF-α as well as the pathological changes of the kidney were observed.Results The expression of kidney tissue＇s α-SMA and TNF-α in Tong Luo Yi Shen Wen Yang formula group was obviously lower than that of the pathological control group （P〈0.01）. The pathological injury of the kidney tissue of Tong Luo Yi Shen Wen Yang formula group was less severe than that of the pathological control group.Conclusion Tong Luo Yi Shen Wen Yang formula has certain therapeutic effect on DN rats,and its probable mechanism is inhibiting the expression of kidney tissue＇s α-SMA and TNF-α,and alleviating the extent of fibrosis of renal interstitium.
