摘要
目的探讨整合素连接激酶(ILK)在病毒性心肌炎(VMC)的发生、发展过程中的作用,及ILK和转化生长因子(TGF)-β1在心肌纤维化形成过程中的作用。方法应用柯萨奇病毒B3(CVB3)感染Balb/c小鼠,建立VMC小鼠模型。实验分为正常对照组(n=30)和病毒组(n=40)。在CVB3感染后第7、14、28、56天应用免疫组织化学方法检测各实验组ILK和TGF-β1的表达,对心肌胶原纤维进行Masson染色和测定血清CK-MB水平,同时分析各实验组不同时间点心肌组织中ILK和TGF-β1阳性表达与心肌胶原纤维Masson染色阳性面积及与血清CK-MB水平的相关性。结果正常对照组无或仅有极少量的ILK和TGF-β1阳性表达,病毒组在病毒感染后各时间点ILK和TGF-β1阳性表达面积均明显增加,与对照组比较差异有统计学意义(P<0.01),且ILK和TGF-β1阳性表达面积存在直线相关性(r=0.970,P<0.01);病毒组小鼠于病毒感染后第14天开始Masson染色阳性面积明显增多(P<0.05);且病毒组小鼠心肌组织中ILK阳性表达面积与心肌胶原纤维Masson染色阳性面积存在直线相关性(r=0.996,P<0.01);心肌组织中ILK的阳性表达面积与血清CK-MB无相关性(r=-0.624,P>0.05)。结论 ILK的异常表达在VMC的发生、发展中起重要作用,在VMC发病进程中ILK参与了心肌纤维化的形成过程。
Objective To explore the role of integrin-linked kinase (ILK) in the process of the occurrence and development of viral myocarditis (VMC) and the effect of ILK, TGF-β1 in myocardial fibrosis. Methods Balb/c mice was infected by coxsackievirus B3 (CVB3) to establish VMC model. The mice were divided into control group (n = 30) and virus group (n = 40). At infected 7, 14, 28, and 56 days, the levels of ILK and TGF-β1 were detected by streptavidin peroxidase conjugate. The area of positive Masson stained myocardium collagen fibers was measured, and the level of serum creatine phosphokinase-MB (CK-MB) was determined. At the same time, the relationship between the expression of ILK and TGF-β1, the positive area of myocardium collagen fibers, and the serum level of CK-MB were analyzed. Results There were no or little expression of ILK and TGF-β1 in control group. Meanwhile, in virus group, the expressions of ILK and TGF-β1 were much higher than that in control group (P 〈 0.01). The expression of ILK and TGF-β1 was positively linear correlated (r = 0.970, P 〈 0.01). The positive Masson stain area of myocardium collagen fibers in virus group was significantly increased than that in control group at 14th day infected CVB3 (P 〈 0.05). The expression of ILK was positively correlated with the positive Masson stain area of myocardium (r = 0.996, P 〈 0.01), but it was not correlated with the level of serum CK-MB (r = -0.624, P 〉 0.05). Conclusions The abnormal expression of ILK is related with the occurrence and development of VMC, and involved in the process of myocardial fibrosis caused by VMC.
