转输树突状细胞疫苗免疫脾淋巴细胞对小鼠骨髓瘤的继承性免疫防治作用 被引量:2

摘要 目的:研究树突状细胞(dendriticcels,DC)类瘤苗免疫小鼠脾淋巴细胞对实验性骨髓瘤的继承性免疫预防与免疫治疗的作用。方法:先前用骨髓DC作为佐剂,研制了DC与肿瘤细胞的融合瘤苗,以及用灭活肿瘤细胞及其裂解产物体外脉冲的DC疫苗,并成功地诱导宿主产生抗肿瘤的免疫保护性反应,且对荷瘤动物具有免疫治疗作用。在此基础上,本研究用DC融合瘤苗及肿瘤抗原脉冲DC疫苗免疫的脾淋巴细胞作为免疫效应细胞,继承性转输到宿主体内,以观察其免疫预防与治疗的效果。结果:继承性免疫预防试验表明,转输免疫脾淋巴细胞后2组宿主均能够抵抗野生性肿瘤细胞的多次攻击,>60d未见皮下移植性肿瘤形成,其60d存活率为100%(5/5),与对照组相比,P<0.01。继承性免疫治疗(adoptiveimmunotherapy,AIT)试验显示:①单用免疫淋巴细胞进行AIT的2组,60d存活率分别为80%(4/5)和60%(3/5),其存活时间也明显延长,P<0.05;②先行化疗后进行AIT,以及术前或术后再行AIT,60d存活率提高到100%(5/5),其存活时间显著延长,P<0.01;AIT后宿主荷瘤生长明显被抑制,且部分荷瘤消退甚或治? Aims: To investigate whether the transfusion of immune spleen lymphocytes with dendritic cells (DC) based cancer vaccines could induce adoptive antitumor immunopreventive (AIP) and immunotherapeutic (AIT) effects.Methods:Previous studies of our laboratory have demonstrated that vaccination of normal and tumor bearing mice with fused cells of bone marrow derived DC with NS 1 myeloma cells and with DCs pulsed with the tumor lysates in vitro can induce host in vivo antitumor immune reponses.In this study,donor immune spleen lymphocytes with the fused or pulsed DCs were used as immunocompetent cells and transfused into recipients of AIP and AIT experiments Results:The AIP experiments indicated that the transfusion of DC induced immunocompetent lymphocytes induced potent immunity,and no tumorigenesis was observed after rechellenges with wild live tumor cells for over 60 days,for which the rate of survival was 100%(5/5, P < 0.01). The AIT experiments showed that ①After AIT only with DC induced immunocompetent cells the 60 day survival rate of tumor bearing mice in two groups was 80% (4/5) and 60% (3/5),respectively.②Using a combination of above mentioned AIT with chemotherapy or surgery,the rate of 60 day survial increased significantly ( P <0.01) and the survival time was extended ( P <0.05).③After alone or combinative AIT,the growth and progression of the model tumor were checked or controlled,and tumor regression occurred in some mice,but it remained to be further confirmed by pathology and molecular biology.Conclusion: The transfusion of immune spleen lymphocytes with the fused or pulsed DCs may induced host to obtain adoptive antitumor immunopreventive and immunotherapeutic effects.
