氧自由基介导血管内皮细胞与白细胞粘附的研究

摘要 目的 探讨氧自由基 H2 O2 介导血管内皮细胞与白细胞粘附以及粘附的机理。方法 实验共分三组。 组 :H2 O2 2 5 0 UM刺激培养至对数生长期的人脐静脉内皮细胞 0 .5、1、2、3和 4小时 ,分别于刺激各时点加入 10 5个白细胞 ,共同孵育 2 0分后 ,镜下计数粘附到内皮细胞上的白细胞数。 组 :抗选择素 P的单克隆抗体与 H2 O2 共同作用内皮细胞 ,刺激时间与观察指标同 组。 组 :免疫细胞化学技术分析受 H2 O2 刺激内皮细胞膜粘附分子—选择素 P的表达。结果  组刺激各时点实验值同对照值比有显著性差异 (P<0 .0 1)。 组实验值同 组相同时点实验值比有显著性差异 , 组内皮细胞受 H2 O2 刺激细胞膜可表达选择素 P。结论  H2 O2 诱导内皮细胞表达选择素 P,后者介导了H2 O2 诱导的血管内皮细胞与白细胞粘附。 Objective To evaluate the adhesion between vascular endothelial cells and leukocytes induced by the oxygen radicals.Methods The subjects were divided into three groups.Group Ⅰ:After the cultured human umbilical venous endothelial cells(HUVEC)were stimulated by H 2O 2 250UM for 0.5,1,2,3 and 4h, 10 5 leukocytes were added to incubate with HUVEC for 20 min,then the adhesive leukocytes were counted under microscope.Group Ⅱ:The HUVEC were stimulated by both anti P selectin mAb and H 2O 2,the method is the same as group Ⅰ.Group Ⅲ:The expression of P selectin on HUVEC induced by H 2O 2 was observed through immunocytochemistry.Results In group Ⅰ,when HUVEC were stimulated by H 2O 2,the adhesive leukocytes increased significantly.The adhesive leukocytes in group Ⅱ decreased siginificantly compared with that in group Ⅰ when HUVEC were stimulated at the same time.In group Ⅲ,H 2O 2 induced expression of P selectin on HUVEC.Conclusion H 2O 2 can stimulate HUVEC to express P selectin which mediates the adhesion between the HUVEC and leukocytes.
