摘要
用自制平板式不锈钢组织粉碎器。直接从手术切除的新鲜人原发性胃癌组织中用异硫氰酸胍-氯化铯超离心法抽提组织总RNA,再用Oligo(dT)纤维素柱分离得到mRNA,以经典的Northern印迹及分子杂交、放射自显影法观察到人原发性胃癌组织中c-H-ras原癌基因的表达强于或等于自身对照的相应正常胃粘膜组织。
This article describes a method to extract total RNA, mRNA and an investigation of the expression of c-Ha-ras protooncogene in human primary gastric cancer. The malignant and the corresponding normal gastric mucosal tissue were obtained from gastrectomy patients, broken into small pieces by a plate form stainless stell splitter, pulverized to tissue powder in a mortor with liquid nitrogen, and the tissue power homogenized in guanindium thiocynate with a polytron. Total RNAs was isolated by guanidium-cesium chloride ultracentrifugal method, mRNAs was then isolated from total RNAs by Oligo(dT)cellulose colum chromatography. Castric total RNAs and mRNAs were analysed by Northern blot method. We found that the expression of c-Ha-ras protooncogene in human gastric cancers was higher than or equal to the corresponding normal gastric mucosal tissues and there was no close relationship between the expression of protooncogene and histology of malignant tissue.
