摘要
[目的]建立一种快速构建多肽基因串联体的方法。[方法]在多肽基因两侧加入同尾酶识别位点,利用同尾酶形成的粘性末端连接后不能被原酶识别与切割的特点,有效构建基因串联体。[结果]利用化学法合成的人表皮生长因子基因,有效构建并获得多肽基因的二联体与三联体。[结论]利用同尾酶技术,可以快速有效地构建基因串联体。
[Objective] The aim was to set up a method for the construction of tandem repeats of polypeptide gene.[Method] The isocaudamer recognition sites were joint with a polypeptide gene on both sides.When the plasmids containing polypeptide gene were cleaved by isocaudamer respectively,the same cohesive ends could connect each other,and the new sequence could not be recut by each of the original isocaudamer enzyme.The gene multimers were effectively constructed.[Result] Using the synthesized human epidermal growth factor gene and this construction strategy of tandem repeats of polypeptide gene,the dimer and trimer of EGF gene were obtained.[Conclusion] A method of construction of tandem repeats of polypeptide gene was established,and the peptide gene repeats would be constructed quickly and efficiently.
