嗅鞘细胞移植对大鼠脊髓损伤的修复作用及机制

摘要 目的探讨嗅鞘细胞(OECs)移植对损伤脊髓的作用及机制。方法取48只成年SD大鼠制备脊髓损伤模型,造模成功后将大鼠随机分为两组各24只,OECs组行OECs移植:取10μl的OECs悬液于脊髓损伤处及损伤段远、近端蛛网膜下腔分别注入8、1、1μl,并各留针2 min,缓慢退针;对照组予生理盐水10μl注入,其他操作同观察组。应用BBB运动功能评分比较两组行为学指标,HE染色、胶原纤维酸性蛋白(GFAP)免疫组化染色分别观察脊髓损伤组织学变化及T9星形胶质细胞增生情况。结果干预后2~8周OECs组运动功能恢复情况、BBB评分均显著高于对照组及干预前,P均<0.01。干预后4周HE染色示对照组脊髓组织有较多空腔,神经纤维及细胞坏死,颜色较暗;而OECs组脊髓组织中无明显空腔,空泡小而少,色泽较亮。GFAP免疫组化染色示两组受损脊髓节段均出现星形胶质细胞反应性增生,但OECs组明显轻于对照组。结论 OECs对损伤脊髓具有修复和保护作用,其机制为促进损伤脊髓的轴突再生,减少星形胶质细胞增生。
