斜带石斑鱼稚鱼和早期幼鱼消化道可培养菌群研究 被引量:3

摘要 利用常规微生物分离鉴定方法结合16S rRNA基因序列分析研究斜带石斑鱼Epinephelus coioides 40d稚鱼和70d早期幼鱼消化道可培养菌群.结果表明,副溶血弧菌Vibrio parahaemolyticus、哈维弧菌Vibrio harveyi、短小芽孢杆菌Bacillus pumilus、鲍氏不动杆菌Acinetobacter baumannii、恶臭假单胞菌Pseudomonas putida、嗜冷杆菌Psychrobact-er sp.和洋葱伯克霍尔德菌Burkholderia cepacia均从稚鱼和早期幼鱼消化道分离.系统进化分析结果表明,斜带石斑鱼消化道细菌可归为3类:γ-变形杆菌纲γ-Proteobacteria、β-变形杆菌纲β-Proteobacteria和芽孢杆菌纲Bacil-li.稚鱼和早期幼鱼消化道的优势菌相似,稚鱼消化道的优势菌为芽孢杆菌Bacillus、嗜冷杆菌Psychrobacter、葡萄球菌Staphylococcus和不动杆菌Acinetobacter,而早期幼鱼消化道的优势菌为芽孢杆菌、不动杆菌、嗜冷杆菌和假单胞菌Pseudomonas.芽孢杆菌为稚鱼与早期幼鱼消化道的最优势菌,分别占可培养细菌总数的78.6%和75.6%,而弧菌在稚鱼和早期幼鱼消化道中的数量均较低,分别仅占消化道可培养细菌总数的2.8%和1.1%.乳酸乳球菌Lactococcus lactis、干酪乳杆菌Lactobacillus casei和屎肠球菌Enterococcus faecium等3种乳酸菌仅见于早期幼鱼消化道. Standard isolation and characterization procedures in combination with 16S rRNA gene sequence analysis were used to study the gut microbiota of 40 d juvenile and 70 d early young grouper. The results showed that seven species existed in the gut of both groups of fish, i. e. Vibrio parahaemolyticus, Vibrio harveyi, Bacillus pumilus,Acinetobacter baumannii, Pseudomonas putida, Psychroacter sp. and Burkholderia cepacia. Phylogenetic analysis showed that bacteria in the gut of grouper could be classified into three groups belonging to γ-proteobacteria, β-proteobacteria and Bacilli class. The predominant bacteria in juvenile and early young grouper were similar, Bacillus, Psychrobacter, Staphylococcus and Acinetobacter were the predominant bacteria in the gut of juvenile fish, while Bacillus,Acinetobacter, Psychrobacter and Staphylococcus in early young fish. Bacillus was the most prevalent bacteria in both groups of fish, comprising 78.6% and 75.6% of the total cultivable gut bacteria respectively, while the number of Vibrio was very low in both groups of fish,comprising only 2. 8% and 1.1% of the total cultivable gut bacteria respectively. Three species of lactic acid bacteria, Lactococcus lactis, Lactobacillus casei and Enterococcus faecium existed only in the gut of early young grouper.
