摘要
目的观察白藜芦醇对糖尿病大鼠血管平滑肌细胞(VSMC)增殖影响及对核因子-κB(NF-κB)的影响。方法采用一次性腹腔注射链脲佐菌素(STZ)55 mg/kg诱发糖尿病大鼠模型,取建模成功大鼠胸主动脉中膜血管平滑肌细胞进行培养,倒置显微镜下观察细胞形态,SMA免疫组化染色鉴定细胞,计数法观察细胞的生长曲线,CCK-8法测定白藜芦醇IC50,共聚焦显微镜下观察白藜芦醇对VSMC中NF-κB核转移的影响。结果与正常组对比,糖尿病组VSMC生长速度快,白藜芦醇能明显抑制糖尿病血管平滑肌细胞增殖,IC50为(38.96±2.14)μmol/L,有剂量依赖性;白藜芦醇能抑制糖尿病VSMC中NF-κB从胞浆转移到胞核。结论白藜芦醇能抑制糖尿病VSMC增殖与NF-κB的核转移。
Objective To investigate the effects of resveratro1(Res) on nuclear factor-κB(NF-κB) activity in proliferation of Diabetes Mellitus rats vascular smooth muscle cells(VSM-C).Methods Diabetes model of rat were established with STZ55mg.kg-1 once through intraperitoneal injection,thoracic aortic vascular smooth muscle cell membrane of diabetes rat were cultured,the cells were observed with inverted microscope,identificated with SMA immunohistochemistry and the cell growth curve were described,the inhibitory rate of 50% concentration were determined by CCK-8,the localization of NF-κB in VSMC with or without breviscapine were estimated by laser confocal microscopy.Results Compared with the normal group,Diabetes VSMC group was grow faster,Res could inhibit DM VSMC proliferation in a dose-dependent manner,IC50 was(38.96±2.14)μmol/L;Res could inhibit NF-κB in DM VSMC transfer from the cytoplasm to the nucleus.Conclusion Res inhibit the proliferation of DM VSMC and NF-κB transfer.
